Analysis of MsTERT Gene Expression Profile in Alfalfa (Medicago sativa) Indicates Their Response to Abiotic Stress and Seed Aging

Seed aging is always taken as a crucial factor for vigor loss due to delayed seed germination and seedling growth, which limits hay production. Many studies have found that telomeres are closely related abiotic stress vigor. However, the molecular mechanism of telomeres’ response stress, vigor, maintenance plant telomere homeostasis still remain unclear. Alfalfa (Medicago sativa) enjoys title “King Forage”, an important protein forage dairy industry planted in world. This comprehensive investigation was performed explore characterization, phylogenetic relationship, gene expression analysis MsTERT under during alfalfa. In this study, identified from ‘Zhongmu 1’ alfalfa genome encoded coding sequence (CDS) 3615 bp length, consisting telomerase- RNA-Binding Domain (RBD) Reverse Transcriptase (RT) domains, 1024 amino acids, isoelectric point 9.58, relative mass 138.94 kD. Subcellular localization showed mainly localized nucleus mitochondria. The results profile observed respond various conditions such salt (100 mmol/L NaCl) drought (20% PEG 6000). Furthermore, exogenous hormones IAA, ABA, GA3 potential affect expression. Additionally, also responded aging. Our revealed marginal but significant association between expression, percentage, suggesting length shortened, decreased with aged.
